During trials on the influence of environmental factors on the parasite of the flat oyster digestive gland, we observed that some experimental conditions including the re-immersion after a several day-exsiccation period could result in a decrease of the parasitism rate.

Etude comparative de différents indices de condition chez l'huître creuse (Crassostrea gigas) ArchiMer

Among the different condition indices, some of them were compared on a population of the japanese oyster, Crassostrea gigas. The follow-up of experimental racks was performed monthly during four years. The use of dry weight in condition indices gave a more precise and more reliable representation for the condition of the oysters than the fresh weight. The sensitivity of these indices, which corresponds to their ability in revealing small changes in the physiological condition, was calculated for each indice. Those exhibiting the best sensitivity were ranked as follow: dry weight on the difference between total weight and shell weight, dry weight on intervalve volume. The lipid content of spawned oocytes of Pecten maximus was studied by image analysis of semi -thin histological sections and a lipid index was defined. The female gametes were obtained by thermic stimulations of mature adults scallops from different populations after out of season conditioning or sampled in situ during the spawning season. After fertilization, the larval development was followed. The results reveal a large variability in lipid content of normal oocytes. The lipid content can be regarded as homogeneous inside a spawning but the heterogeneity is greater for wild scallops than for conditioned ones. Polyploidy in the Manila clam Ruditapes philippinarum. II. Chemical induction of tetraploid embryos ArchiMer

Tetraploidy was induced in embryos of Ruditapes philippinarum by treating the eggs with cytochalasin B. (CB). Two treatment times were found to be effective in inducing tetraplidy : 0 to 10 minutes after insemination (before formation of the first polar body) and 45 minutes after insemination (just before the first cleavage). Treatments, started 5 and 45 minutes after insemination, produced 64.4 ± 7.8% and 28.3 ± 2.3% tetraploid embrios, respectively, after 6 hours (on the average over 3 experiments). However, no tetraploids were detected in 4-month-old spat. Sperm entry delayed by the CB could be involved in induction of tetraploidy by early treatment.

Polyploidy in the Manila clam Ruditapes philippinarum - 1 - Chemical induction and larval performances of triploids ArchiMer

An effective treatment using catochalasin B (1 mg CB/ml dimethylsulfoxyde in 1 litre of sea water) was developed to induce triploidy in the Manila clam Ruditapes philippinurum (Adams and Reeve). The percentage of induced triploidy was assessed by karyological examination of embryos and spat. The treatment was found to be optimal between 20 to 35 minutes post-insemination exposure to CB, at 25'C, and produced 75.8 ± 5.7% triploid embryos on the average over four experiments. Growth and survival of larvae to metamorphosis were recorded for two different rearings each split into two replicates. The sexual maturation in bivalve molluscs causes decreased growth and sometimes increased mortalities. These drawbacks can be avoided by genetic sterilization (triploidy). La conservation des échantillons a souvent constitué un problème tant pour les chimistes que pour les biologistes. Si les chimistes semblent satisfaits de leurs techniques de conservation, les biologistes quant à eux sont indécis du fait que l'action toxique se modifie au cours du temps et selon la technique employée (réfrigération, congélation, lyophilisation, conservation en azote liquide à -196°C). Le présent rapport constitue une étude préliminaire qui, si elle est poursuivie, pourrait permettre de déterminer le mode de conservation le plus approprié pour chaque type d'échantillons.

Analysis of shell colour determinism in the Manila clam Ruditapes philippinarum ArchiMer

The Manila clam presents a large polymorphism for shell colours and patterns. No experiment has been conducted to study the determinism of such a polymorphism which has been well known for a long time. The aim of this study was firstly to determine the genetic variation for shell colour and secondly to produce some homozygous lines for the shell colouring. These lines could then be used as controls for laboratory experiments. The colouring is defined by three characters: asymmetry, ornamentation and hue. Bacterial problems associated with scallop Pecten maximus larval culture ArchiMer

Scallop Pecten maximus larvae have been cultured at the Argenton and Tinduff (Brittany, France) hatcheries with antibiotic treatment (chloramphenicol at 8 ppm) for 15 yr. Without treatment, outbreak of disease has normally occurred between Day 12 and Day 19 or sometimes earlier. A bacteriological study of larvae reared with and without antibiotic was performed over a 4 yr period. Among the collected strains, 2 clusters (C and F) of vibrios were present at high densities only in larvae cultured without treatment. A short description of the disease causing organisms affecting bivalves is presented in this document, including the diagnosis, the oustanding research and bivalve husbandry. Mollusc populations may be safe from disease epizootics if an early prevention of disease is coupled with a strict regulatory system on transfer (according to what happens in agriculture). The species to culture may be thoroughly selected and risky populations moved. Imported stocks are strictly controlled. The researchcs's target is to improve the diagnosis tools and try to find resistant mollusc strains. Epidemiological surveys are also necessary to control diseases.

Impact of the oyster Crassostrea gigas on a microbial community in Atlantic coastal ponds near La Rochelle ArchiMer

To assess the in situ impact of oysters Crassostrea gigas on planktonic protist and bacteria communities and the potential contribution of protozoa to their food resource intake, the abundance and the diversity of protists and bacteria were followed in 2 Atlantic coastal ponds, with and without oysters. The protist biomass in such ponds was high, with a maximum in spring of 982 mug C l(-1) and a minimum in winter of 179 pg C l(-1). Whatever the season, the presence of oysters (20 m(-2) corresponding to an average of 23 mg dry weight m(-2)) induced a significant decrease in >5 pm protist abundance. Evidence for interspecies transmission of oyster herpesvirus in marine bivalves ArchiMer

Since 1991, numerous herpesvirus infections associated with high mortality have been reported around the world in various marine bivalve species. In order to determine whether these infections are due to ostreid herpesvirus-l (OsHV1), a previously characterized pathogen of the Japanese oyster (Crassostrea gigas), PCR analysis was carried out on 30 samples of larvae collected from four bivalve species (C. gigas, Ostrea edulis, Ruditapes decussatus and Ruditapes philippinarum), most exhibiting mortality prior to collection. All samples were shown to be infected by OsHV1.
